      【目的】克隆甘蔗内生固氮菌变栖克雷伯氏菌(Klebsiella variicola)DX120E nifH基因,并对该基因的生物信息学进行分析,为探讨甘蔗内生固氮菌DX120E的固氮分子机制提供理论依据。【方法】根据NCBI上其他固氮菌株的nifH序列设计引物,内生固氮菌Klebsiella variicola DX120E菌株为材料进行PCR扩增,运用生物信息学方法对其核苷酸序列、编码的氨基酸序列进行分析以及对其蛋白结构进行预测。【结果】甘蔗内生固氮菌Klebsiella variicola DX120E nifH基因的ORF为882 bp,GenBank登录号为KF732646.1,与Klebsiella pneumoniae 342、Klebsiella variicola At-22等6种固氮菌的nifH 基因同源性高达95%-99%。【结论】甘蔗内生固氮菌Klebsiella variicola DX120E的nifH基因与克雷伯氏菌属其他种的nifH基因的同源性很高, 而与非克雷伯氏菌属的固氮菌的亲缘关系相对较远。

      【Objective】 This study aimed to clone nifH gene of Klebsiella variicola DX120E isolated from sugarcane and then made bioinformatics analysis in order to provide references for understanding the molecular mechanisms of nitrogen fixation by the endogenous nitrogen-fixing bacteria DX120E in sugarcane. 【Method】According to the nifH primers of other nitrogen-fixing bacteria strains registered in the NCBI, the primer was designed and PCR amplification was done for the endogenous nitrogen-fixing bacteria strain Klebsiella variicola DX120E from sugarcane. Bioinformatics methods was used to analyze its nucleotide sequence, amino acid sequence encoded by nifH gene, and predicted its protein structure.【Results】Sugarcane endogenous nitrogen-fixing bacteria strain Klebsiella variicola DX120E nifH gene had an ORF which length was 882 bp. The gene has been registered in GenBank with accession number KF732646.1. The homology of nifH gene in Klebsiella variicola DX120E with other 6 nitrogen-fixing bacteria including Klebsiella pneumoniae 342, Klebsiella variicola At-22 were between 95%- 99%. 【Conclusion】We had cloned the nifH gene in sugarcane endogenous nitrogen-fixing bacteria Klebsiella variicola DX120E and it was showed that the nifH gene has high homology with other nitrogen-fixing species of Klebsiella, but relatively lower with non- Klebsiella species.
